    PRODUCTION AND CHARACTERIZATION OF CRUDE INTRACELLULER PHYTASE FROM RECOMBINANT BACTERIA pEAS1AMP

    This research was aimed at producing a crude intracellular phytase characterized from recombinant bacteria. The recombinant bacteria (pEAS1AMP) was produced by way of transforming pET-22b(+) +pEAS1 into competent E. coli BL21 and E. coli BL21(DE3) cells. Crude intracellular phytase production was induced using 1,5 mM Isopropyl-β-D-thiogalactopyranosid (IPTG). Recombinant bacteria product and enzyme activity test followed the Sajidan method. E. coli BL21(+)pEAS1 and E. coli BL21 (DE3)(+)pEAS1 recombinant bacteria showed growth after 20 hours and 10 hours of transformation. Phytase activity of E. coli BL21 (DE3)(+)+pEAS1 showed higher than those of E. coli BL21(+)+pEAS1. Crude intracellular phytase of pEAS1AMP recombinant bacteria has an optimum activity at pH 5, 40o C, incubation period of 60 minutes, substrate concentration of 2%, molecular weight (MW) of 47.3 kDa, Km = 15.91 υM and Vm = 2.41 υM/second.     In Vitro Stability of Phytase from Recombinant Bacteria E. Coli BL21 (DE3) EAS1-AMP

    The objective of the research was to inquire the Km, Vm, activity, intracellular phytase stability exposed to pH variation, temperature variation and protease (pepsin and pancreas) in vitro. The phytase was produced from recombinant bacteria E. coli BL21(DE3) EAS1-AMP using 1.5 mM IPTG as inducer. Intracellular enzyme was extracted via freeze shock and centrifugation. Pure enzyme was acquired through NI-NTA agarose column. The enzyme was then tested for Km, Vm, phytase activity and stability against pH, temperature and protease. Treatment levels for stability against protease were P0: without protease, P1: addition of pepsin, P2: addition of pepsin and pancreas, and the data were statistically analyzed using analysis of variance of one-way Completely Randomized Design. Crude intracellular phytase had Vm 6.39 υM/sec, Km 34.82 υM, and 277 units activity. Intracellular phytas was stable at pH 4–6 and 0–550 C. Protease level influenced the activity of intracellular phytase (P<0.05). Intracellular phytase was stable against pepsin but not pancreas.     Study on effect of variations of meta-kaolin addition on Self-Compacting parameter of High Strength Concrete

    High strength self-compacting concrete (HSSCC) is one of the concrete innovations that have a high quality standard. This type of concrete has a high compressive strength and can compact without tools. Meta-kaolin (MK) is used in this research as an addition (admixture) to increase the quality of HSSCC and to meet criteria of SCC. Meta-kaolin (MK) is made from kaolin which a white powder is measuring 0.5 to 5 micron that then heated at 500– 900oC. Parameters that are studied in this research are setting time, workability, and the compressive strength of HSSCC. Final results of the research shows that the addition of MK decrease workability of HSSCC. Only MK addition at 5%, 7.5%, and 10% fulfils filling ability and passing ability parameter of SCC, otherwise the compressive strength increases linearly

    Serum Level Changes of Neurotrophin-3 After Performing Diabetic Foot Exercise in Diabetic Neuropathy

    The research aimed to determine the pattern of changes in serum levels of NT-3 in the improvement of diabetic neuropathy, after doing diabetic foot exercise. A true experimental study with randomaized pre – post test control trial. A total of 36 subjects meeting the inclusion and exclusion criteria were included in the exercise group or the control one with age matched systematic random sampling method. Exercise group had a significant improvement on the score of ABI (p.0.002), systolic blood pressure (p.0.014), diastolic blood pressure (p.0.055), DNS (p.0.01), DNE (p.0.001) and increased of serum level of NT-3 (p.0.049). Control group had result respectively on ABI (p.0.131), systolic blood pressure (p.0.668), diastolic blood pressure (p.0.216), DNS (p.1.00), DNE (p.0.543), and increase of NT-3 (p.0.264). The comparation results of the two groups had a significant different on the score of ABI (p.0.01), systolic blood pressure (p.0.01), diastolic blood pressure (p.0.01), DNS (p.0.01), DNE (p.0.01), and increased of NT-3 (p.0.01). Diabetic foot exercise had a peripheral affect on a clinically significant improvement based on ABI scores, systolic and diastolic blood pressure, DNS and DNE, and increase of serum level of NT-3

    Congener specific analysis of polychlorinated terphenyls

    In order to identify and to quantify polychlorinated terphenyls (PCT) in environmental matrices, the chro- matographic behavior of coplanar and non-coplanar congeners was evaluated. A mixture of 16 single PCT congeners was used for method development. Four of these compounds were synthesized for the first time by SUZUKI-coupling reaction. These were p-PCT (2,200 ,6,600 -tetrachloro-, 20 ,3,300 ,4,400 ,50 ,-hexachloro-, 20 ,3,300 ,5,50 ,500 -hexachloro-) and m-PCT (2,200 ,3,300 ,5,500 -hexachloro-). They were characterized by NMR ( 1 H, 13 C) spectroscopy. By means of the new column chromatographic clean-up reported here, a good matrix removal and the separation of the coplanar PCT congeners from the non-coplanar ones was obtained. The recovery rates for all congeners were good for the PCT in different test matrices like fat, charcoal, and soil. The quality of the clean-up, the separation and the recovery rates were determined by GC/MS analysis. The method was applied for the first time to a real sample from a fire accident, where different PCT, obviously formed during the combustion process, were found. The conclusion is drawn that this method is suitable for the analysis of PCT in different environmental samples

The bidders usually consider various aspects, including internal, external and environment, with aim to win the bidding competition, and provide maximum project performance.The literature review method is used to create ideas, and synthesize the related researches which have been done previously. This paper aims to develop a framework that can be used for evaluation in the early stages of project selection, based on theriview of various literature related to bidding strategies and project performance. The result from the literature review concerning are presented, and a conceptual framework of bidding strategy model is developed

    This study aims to explore the process of internalizing the values of national character through PAI subjects which include context, input, process, and the product internalization of the values of nation character, through the lesson of PAI learners. By using a qualitative approach and CIPP (Context, Input, Process, Product) methods the study found four findings. (1) In the context,the strategy of planting the values of national character, through religious subjects at high school students conducted by school policy, school system, quality of facilities and infrastructure, as well as the culture at any educational institution. (2) in the input, the internalization of the values of the nation's character do subjects PAI through a qualification and competencies of teachers, the input facilities and infrastructure, and qualified students. (3) the process of internalizing the values of character, through subjects PAI done through a curriculum syllabus and lesson plan, and extracurricular activities intracurricular. (4) products resulting from the internalization of the values of character, through Islamic religious education are that learners have a competence in the field of academic and nationality character at the same time

    Currently, there is a tight competition in service industry, especially in higher education or university. The administrators or universities have to understand deeply their students and try to fulfill their needs well to make them satisfied and tell positive word-of-mouth about the university. Therefore, the understanding of student satisfaction and other factors determining student satisfaction, such as service quality, reputation, and service value of the university, become very important. The objectives of this study are to know whether there are positive effects of service quality and reputation on service value. Furthermore, this study also try to verify whether there are positive effects of service quality and value on student satisfaction. Results indicate that most of the empirical data support that there are positive effects of service quality and reputation on service value. Likewise, this study proves that there is a positive effect of service value on student satisfaction. However, this study also find that there is not a positive effect of service quality on student satisfaction
